Sulfanilic acid reacts with sodium nitrate to form diazotized sulfanilic acid. In the presence of Dimethylsulfoxide, total bilirubin  reacts  with  diazotized  sulfanilic  acid  to  form azobilirubin. In the absence of Dimethylsulfoxide, only the direct bilirubin reacts to give azobilirubin
